Cognitive and Neuroprotective

P21 (P021)

A small peptide modeled on CNTF (a nerve growth factor). It pushes the brain to grow new neurons and make more BDNF, studied for memory and Alzheimer’s in animals. Usually taken as a nasal spray so it reaches the brain directly. Cumulative; effects build over weeks.

Overview

A small peptide modeled on CNTF (a nerve growth factor).

It pushes the brain to grow new neurons and make more BDNF, studied for memory and Alzheimer’s in animals.

Usually taken as a nasal spray so it reaches the brain directly.

Cumulative; effects build over weeks.

What It Does

P21 (P021) is a CNTF-derived neurotrophic tetrapeptide mimetic.

It promotes neurogenesis, raises BDNF, inhibits leptin-pathway-linked tau pathology, and shows disease-modifying effects in Alzheimer’s models.

Intranasal delivery exploits nose-to-brain transport, bypassing the blood-brain barrier.

No completed human trials.

Reconstitution Calculator

Enter the vial amount and diluent volume to calculate concentration. If you already have a target amount, you can also convert it to liquid volume and syringe markings. This tool does not recommend a target amount.

Calculated concentration—Enter both values above.
Optional conversion

Enter a target amount you already have and select the syringe capacity.

Calculated draw—Enter concentration inputs and a target amount.

Mathematical reference only. The calculator does not determine an appropriate target amount. Syringe capacity only changes the maximum volume that fits.

Protocol

PhaseFrequencyAmountNotes
Weeks 1–6Daily250–500 mcg SubQ or 500 mcg–1 mg intranasalIntranasal is the common route. Effects are cumulative.
Off2–4 weeks–Cycle 4–8 weeks on / 2–4 weeks off. Caution: no completed human trials, doses are user/animal-derived.
Timing

daytime; neurogenic effects build over weeks, do not expect immediate changes. Route: intranasal gives direct nose-to-brain delivery; SubQ also used.

Population Considerations

Unisex experimental neurogenic peptide, animal data only, no sex-specific dosing. Avoid in pregnancy.

Handling & Stacking

Injection site
Intranasal spray (common) or SubQ abdomen. Rotate.
Storage
  • Freeze-dried: -4°F.
  • Reconstituted: 36–46°F.
  • Best used within ~3–4 weeks, kept cold (community estimate, no published data); that is close to the 28-day sterility limit, so use whichever comes first.
Side effects
Generally mild in reports, nasal irritation (intranasal), mild headache. Human profile not formally characterized.
What to expect
  • Gradual memory/cognitive support over weeks if effective; cumulative.
  • Strongest rationale is in age-related cognitive decline (animal models).
  • Supply math 5mg → 2 vials (6-wk @ 250 mcg/day SubQ; more if intranasal mg-dosing)
Pairs well with
Semax / Selank (cognition + mood); Cerebrolysin (neurotrophic synergy); NAD+
Avoid
Active malignancy (growth-factor signaling, theoretical). Pregnancy

Safety & Patient Education

Contraindications
Pregnancy and lactation Active malignancy (theoretical growth-factor concern) Hypersensitivity to the peptide
Warnings
Neurotrophic/growth signaling, theoretical caution with malignancy Quality varies, request a COA Effects are cumulative, not an acute nootropic
Drug interactions
No well-characterized interactions Stacks with other nootropic peptides, unproven combinations
Labs
None established; cognitive tracking if used for that purpose
Stop criteria
Persistent nasal irritation (intranasal) or headache Any unexpected systemic reaction Pregnancy
Patient education
Cumulative neurogenic peptide, give it weeks, not days Intranasal delivery targets the brain directly Request a COA; source quality matters
